Research interests
Our group has a longstanding interest in the role of viruses in human leukaemia and lymphoma.
Much of our research is focused on Hodgkin lymphoma, one of the commonest malignancies affecting young adults. A proportion of cases of Hodgkin lymphoma, around one third in developed countries, are associated with the Epstein-Barr virus (EBV) and this association is thought to be causal. Epidemiological data suggest that non-EBV-associated Hodgkin lymphoma may be associated with delayed exposure to a common infectious agent(s) and we believe that another virus may be involved in the pathogenesis of this group of cases.
We are currently using next generation sequencing to search for novel viral sequences in tumour cells enriched from biopsy samples from these cases. Recent work from our group, including collaborative Genome Wide Association Studies, provides clear evidence that host genetics influences risk of Hodgkin lymphoma. In particular, HLA genotype modifies disease risk. Genetic risk factors for EBV-associated and non-associated Hodgkin lymphoma are largely distinct, providing further evidence that these two forms of the disease have different natural histories.
Ongoing work is exploring the biology underlying these associations and determining their clinical significance. We are also investigating biomarkers that appear promising both in the early diagnosis of Hodgkin lymphoma and for assessing response to treatment.
Other interests include:
- virus discovery in a broader range of diseases of human and animals including canine lymphoma;
- the significance of germline transmitted, chromosomally integrated human herpesvirus-6;
- non-viral aspects of the pathogenesis of Hodgkin lymphoma.
